Most mechanistic studies in liver cancer are performed using animal models, but important differences exist with human biology. In this study, Sun et al. establish human liver organoids using directly reprogrammed human hepatocytes. The newly generated hepatocytic organoids were then used to model oncogenic events involved in hepatocellular carcinoma and cholangiocarcinoma tumor initiation.
